摘 要:目的测定附子中乌头碱、新乌头碱和次乌头碱的含量。方法采用反相高效液相色谱(PR-HPLC)法,使用Ulti-mateTM XB-C18柱(4.6 mm×250 mm,5μm);流动相为乙腈-0.2%冰醋酸(浓氨水调节pH值为7.29);检测波长:230nm;流速1.0 ml.min-1;柱温为35℃。结果乌头碱、新乌头碱、次乌头碱的回归方程分别为:Y=31.258X+5.794,r=0.999 9;Y=17.418X+5.236 9,r=0.999 8;Y=18.989X+6.186 6,r=0.999 7。三者的线性范围分别为:3.3125~106μg/ml;0.735 7~23.541 9μg/ml;1.966 1~62.916 4μg/ml。结论该方法简便、准确、分离效果好、灵敏度高,可用于附子的质量控制。Objective To develop a high performance liquid chromatography (HPLC) method for the determination of three principal alkaloids in aconite root. Methods (4.6mm × 250mm, 5μm)column by isocratic elution using Acetonitrile -0.2% Glacial acetic acid( adjusted to pH 7.29 with concentrated ammonia). Results The average recovery of the three alkaloids were analyzed simultaneously with UhimateTM XB - C18 rates obtained were in the range of 98.93 - 100.5% for RSD below 2.0%. The linear ranges of aconitine; hypaconitine; mesaconitine were respectively 3. 312 5 - 106 μg/ml, 0. 7357 - 23. 5419 μg/ml, 1. 9661 - 62. 9164μg/ml ( r = 0.999 9, r = 0. 999 8, r = 0. 999 7 ). Conclusion The contents of alkaloids in aconite root varied significantly from species ; hence quality control of aconite root is very necessary.
