摘 要:目的观察田基黄不同提取物的含药血清体外抗乙肝和抗肝癌的作用。方法采用酶链免疫法,观察田基黄不同提取物的含药血清对2215细胞分泌HBeAg和HBsAg的抑制作用。采用MTT技术,观察田基黄不同提取物的含药血清对肝癌细胞BEL-7404增殖的抑制作用。结果当含药血清浓度为5%时,田基黄乙醇总提取物和正丁醇提取物对HBeAg和HBsAg呈现较好抑制作用,抑制率分别为34.84%和57.95%,以及76.02%和53.15%(P<0.05);当含药血清浓度为20%时,醋酸乙酯提取物对HBeAg和HBsAg呈现较好的抑制作用,抑制率分别为79.79%和57.22%(P<0.05);水提取物3种浓度的含药血清对HBeAg和HBsAg都有较好的抑制作用,抑制率分别达到了70%和30%以上(P<0.05)。田基黄各提取物对肝癌BEL-7404细胞都有一定的抑制作用,该抑制作用呈明显的量效关系,当含药血清浓度为20%时,乙醇总提取物、醋酸乙酯提取物、正丁醇提取物和水提取物的抑制率分别为29.74%,53.80%,40.79%和54.24%(P<0.01)。结论田基黄不同提取物含药血清能抑制2215细胞分泌HBeAg和HBsAg,并且能抑制肝癌细胞BEL-7404的增殖。其抗乙肝的有效成分主要分布在田基黄的水提取物中,而抗肝癌的有效成分则均衡分布于各种极性部位。Objective To observe the anti - hepatitis B and anti - hepatoma effect of different extracts in serums of Hypericum japonicum ThuMb. in vitro. Methods Enzyme linked immune response was used to investigate the effect of the different extracts in serums of Hypericum japonicum ThuMb. on HBeAg and HBsAg excreted by 2215 cell in vitro. MTT technique was used to investigate the effect of the different extracts in serums of Hypericum japonicum Thunb. on the growth of hepatoma BEL - 7404 cell in vitro. Results EtOH and BuOH extracts showed better inhibitory rates on HBeAg and HBsAg as 34.84% ,57.95% and 76.02%, 53.15% (P 〈 0.05)in 5% serum respectively. EtOAc extracts showed better inhibitory rates on HBeAg and HBsAg as 79.79% and 57.22% (P 〈 0.05)in 20% serum respectively. H20 extracts had stable inhibitory rates on HBeAg and HBsAg in the three different concentration of serum and the inhibitory rates were all more than 70% and 30% respectively. All extracts showed the inhibitory activity on the growth of hepatoma BEL - 7404 cell in vitro and there was obvious dose - effect relationship. The inhibitory rates of EtOH,EtOAc,BuOH and H2O extracts in 20% serum were 29.74% ,53.80% ,40.79% and 54.24% ,respectively ( P 〈 0.01 ). Conclusion The different extracts of Hypericum japonicum Thunb. in serum have the inhibitory effect on HBeAg and HBsAg excreted by 2215 cell and on the growth of hepatoma BEL - 7404 cell in vitro. The stable active compounds of anti - Hepatitis B mainly distribute in H20 extracts and the active compounds of anti - hepatoma evenly distribute in each extracts.
