摘  要:目的研究毛冬青提取物对大鼠脑缺血再灌注损伤后TNF-α及Caspase-3表达的影响。方法将健康SD大鼠随机分为:假手术组、模型组、毛冬青三个剂量组。采用线栓法栓塞大鼠右侧大脑中动脉1 h,再灌注24 h,制备大鼠脑缺血再灌注模型。用免疫组化SABC法染色,观察25,50,100 mg.kg-1毛冬青提取物对脑缺血再灌注大鼠的脑组织肿瘤坏死因子-α(TNF-α)及半胱氨酸蛋白酶-3(Caspase-3)表达的影响。结果与假手术组比,CIRI模型组脑组织TNF-α及Caspase-3的表达增加;与CIRI模型组比,各剂量毛冬青提取物均能使脑组织TNF-α及Caspase-3的表达下降。结论毛冬青提取物能减少脑缺血再灌注损伤后TNF-α及Caspase-3表达的表达,并对脑组织产生一定的保护作用。Objective To study the influence of expression of TNF -α and Caspase - 3 of Radix ilecis pubescens extract on cerebral ischemia reperfusion injury in rats. Methods The healthy SD rates were randomly divided into the sham - operative group, model group and Radix ilecis pubescens extract groups. Focal cerebral ischemia reperfusion model was established in rats through occluding the middle cerebral artery on one side for 1 hour, and then repeffusion for 24 hours to prepare the rots'cerebral ischemia reperfusion model. Use the immunohistochemisty SABC to stain, and observe the influence of Radix ilecis pubescens extract at doses of 25, 50, 100 mg × kg^ - 1 on the expression of TNF -α and Caspase - 3. Results Compared with the sham - operative group: the expression of TNF- α and Caspase -3 of model group increased. Compared with the CIRI group: the Radix ilecis pubescens extract groups can decrease the expression of TNF - α and Caspase - 3. Conclusions Radix ilecis pubescens extract can decrease the expression of TNF - α and Caspase - 3 after focal cerebral isehemia reperfusion injury, and can protect cerebral.
