摘 要:采用真菌的28SrDNA的特异性引物对千山油松阔叶混交林中乔木层、灌木层和草本层优势植被根区土壤中提取的真菌总DNA进行PCR扩增,并通过DGGE技术对PCR产物进行分析,结果表明:乔木层针叶树(油松、红松)根区土壤真菌的多样性最低,草本层植被根区土壤真菌的多样性最高。乔木层阔叶树根区土壤真菌群落结构与灌木层相似,草本层与二者差异较大。An experiment was conducted to study the structure of soil fungal community in root zone of dominant vegetations of arbor, shrub and herb layers in Pinus tabulaeformis broad-leaved mixed forest in Qianshan by PCR-DGGE. Partial genes from soil fungal community DNA were amplified using primers within 28S rDNA genes in the fungi. The diversity of PCR- amplified products was detected by denaturing gradient gel electrophoresis (DGGE). Results showed that the soil fungal community of conifer in arbor layer had the lowest diversity, while the herb layer had the highest diversity. The PCA analysis indicated that the structure of soil fungal community in root zone of broad-leaved trees in the arbor layer were similar to that in the shrub layer, and the herb layer had a great difference compared with the arbor and shrub layers.
