摘 要:目的:探讨宫颈鳞状细胞癌组织中DNA甲基化转移酶(DNMT1)mRNA的表达和上皮钙粘附素(E-cadherin)的甲基化状况,分析其与宫颈鳞状细胞癌发生发展的关系。方法:应用原位杂交技术检测20例正常宫颈、60例CIN(CINⅠ、CINⅡ、CINⅢ各20例)和40例宫颈鳞状细胞癌组织中DNMT1 mRNA的表达情况;利用甲基化特异性PCR(methylation specific polymerase chain reaction,MSP)方法检测E-cad基因启动子区甲基化状况,分析宫颈鳞状细胞癌中DNMT1 mRNA的表达与E-cad启动子区甲基化状况及2者之间的相关性。结果:DNMT1 mRNA在正常宫颈、CIN和宫颈鳞状细胞癌组织中表达逐渐增加,差异有统计学意义(P<0.05);DNMT1 mRNA在CINⅠ、CINⅡ、CINⅢ组织中表达的差异有统计学意义(P<0.05)。E-cad启动子甲基化率在正常宫颈、CIN、宫颈鳞状细胞癌组织中依次升高,差异有统计学意义(P<0.05)。E-cad启动子甲基化与DNMT1 mRNA的表达呈正相关(r=0.401,P<0.05)。结论:DNMT1 mRNA过表达和E-cad启动子异常甲基化与子宫颈鳞状细胞癌的发生发展关系密切;E-cad启动子甲基化过程可能是在DNMT1的催化作用下完成的。Aim: To study the expression of DNA methyhransferase 1 ( DNMT1 ) mRNA and CpG islands methylation status of E-cadhcrin's promoter region in normal uterine cervix, CIN tissues and cervical cancer, as well as the correlations with clinicopathologic characteristics, and to analyze the relationships between them. Methods:Expression of DNMT1 mR- NA was detected by in situ hybridization in 20 cases nomal cervical tissues, 60 cases CIN( CIN Ⅰ ,CIN Ⅱ ,CIN Ⅲ ,20 cases respectively) ;MSP was to determine the methylation status of CpG island in promoter region of E-cad. The relationship be- tween them was statistically analyzed. Results:DNMT1 mRNA expression in normal uterine cervix,CIN tissues and cervical cancer increased gradually. There is a significant difference among them( P 〈 0.05) ;DNMT1 mRNA expression in CIN Ⅰ , CIN Ⅱ , CIN Ⅲ was significantly different( P 〈 0.05 ). The frequency of CpG islands methylation status of E-cad promoter region in normal uterine cervix, CIN tissues and cervical cancer increased gradually, and a statistical significance was obtained (P 〈 0.05). The expression of DNMTI mRNA was significantly correlated with CpG islands methylation status of E-cad promoter region ( r = 0. 401, P 〈 0.05 ). Conclusion : DNMT1 overexpression and E-cad promoter abnormal methylation may be correlated with cervical cancer closely; DNMT1 may play an important role in inactivation of E-cad gene.
