摘　　要：目的探讨KLOTHO基因第四外显子C-1818T多态性与中国苏皖地区汉族人群急性心肌梗（acute myocradial infarction，AMI）的可能关系。方法入选229例AMI患者（AMI组）和235例经相关检查排除冠心病（CAD）者为对照组，采用聚合酶链反应（polymerase chain reaction，PCR）及基因芯片（gene microarray）技术检测KLOTHO基因C-1818T多态性的基因型，同时测定甘油三酯、低密度脂蛋白、高密度脂蛋白和血糖等临床生化指标。结果与对照组相比，AMI组老年人（≥60岁）和男性患者的CT基因型频率均明显升高（P值分别为0．042和0．039），在调整多种危险因素并经多元Logistic回归分析后，KLOTHO基因CT基因型与老年人和男性AMI无显著关联性。结论KLDTHO基因C-1818T多态性CT基因型可能是中国苏皖地区汉族人群AMI的易感基因型，在存在多种动脉粥样硬化相关危险因素的情况下，携带该基因型的老年人和男性较易发生AMI。Objective To investigate the association between the KLO/THO gene C - 1818T polymorphism in exon 4 and acute myocradial infarction（AMI）in the Chinese Han population of Suwan region. Mothods 229 patients with AMI（AMI group） and 235 control patients without coronary artery disease through coherence checks（control group）were selected and their KIE/THO gene C- 1818T polymorphism were measured by polymerase chain reaetion（PCR）and gene mieroarray. The level of triglyeride, low- density lipopmtein,high - density lipoprotein and glucose were also measured simultaneously by automatic biochemistry analyzer.Results Compared with control group,the frequencies of CT genotype in elderly（ age≥ 60 years）or in male was higher significantly in the AMI group,P value was 0.042 and 0.039 respectively. Furthermore,after adjustment for conventional risk factors by multiple logistic regression analysis,the carrier prevalence of the CT genetype in elderly or in male was not differ between AMI group and control group. Conclusion The CT genetype of the KLOTHO gene C- 1818T polymorphism may be associated with the susceptibihty to AMI in the elderly or male in the Chinese Han population of Suwan region. If had correlated risk factors of atberoselerosis, AM/may be happen in elderly or male.
