摘  要:为了探索刚地弓形虫代谢分泌抗原(E/SA)对山羊的免疫保护机制,将山羊随机分为6组,即E/SA组、E/SA+CpG组(未乳化)、E/SA+CpG组、E/SA+IL-2组、E/SA+IL-2+CpG组、对照组,每组3只,分别于免疫前、免疫后第2周、免疫后第4周、感染后第1周及感染后第2周采集山羊血清,用ELISA法检测IFN-γ、TNF-α、IL-2、IL-4的表达水平。结果显示,各免疫组免疫后IFN-γ、TNF-α、IL-2、IL-4的含量分别为175.40~215.60、89.95~253.80、230.40~301.50、39.20~54.20pg/mL,而免疫前分别为14.00~18.75、30.91~42.20、10.75~18.30、20.40~24.60pg/mL,对照组分别为16.50~36.56、37.75~58.20、11.50~21.75、21.61~27.60pg/mL,免疫组在免疫后体内IFN-γ、TNF-α、IL-2、IL-4的含量明显升高,均显著高于对照组(P<0.05)。表明,E/SA可引起IFN-γ、TNF-α、IL-2、IL-4水平的升高,说明E/SA免疫后可引起山羊较强的细胞免疫和体液免疫应答。To elucidate the effect of Toxoplasma gondii excreted/secreted antigens on goat's immune function,goats were randomly divided into 5 immune groups of E/SA, E/SA+CpG (without emulsification),E/SA+CpG,E/SA+IL-2,E/SA+IL-2+CpG,and the control group with 3 each. Sera were collected and the level of IFN-γ,TNF-α,IL-2 and IL-4 was analyzed by ELISA before immunization,on week 2 post-immunization,on week 4 post-immunization, on week 1 post-infection and on week 2 post-infection, respectively. The levels of IFN-γ, TNF-α, IL-2 and IL-4 were 175.40 - 215.60,89.95 - 253.80,230.40 - 301.50 and 39.20-54.20 pg/mL after immunization,respectively,but were 14.00-18.75,30.91-42.20, 10. 75-18.30,20.40-24.60 pg/mL pre-immunization in the 5 immune groups. The levels were 16.50- 36.56,37.75-58.20,11.50-21. 75 and 21.61-27.60 pg/mL in the control group. The levels of IFN-γ, TNF-α,IL-2,IL-4 in the 5 immune groups were significantly higher than that in the control group(P〈0.05) post-immunization. The results that E/SA elevated the levels of IFN-γ,TNF-α,IL-2 and IL-4 showed that E/SA could result quickly in cellular immune response and humoral immune response.
