摘  要:用分离自乳腺炎病牛乳样中的金黄色葡萄球菌感染BALB/c小鼠,复制出了乳牛乳腺炎模型。将90只泌乳BALB/c小鼠随机分为健康组、模型组、治疗组,每组30只。于攻菌后不同时间点收集乳腺,用HE染色法观察乳腺组织的病理变化;用甲苯胺蓝染色方法观察其肥大细胞形态特征;用免疫组织化学染色法检测乳腺中iNOS和3-NT的表达;用酶联免疫吸附试验(ELISA)检测乳腺组织中TNF-α的水平。结果显示,在攻菌后第24h形成标准乳腺炎模型,出现了大量中性粒细胞浸润、充血、腺上皮细胞坏死等病理变化,治疗组中性粒细胞减少,充血不明显,病理变化减轻。与模型组比较,治疗组乳腺肥大细胞数量及脱颗粒现象减少,iNOS和3-NT阳性表达降低,TNF-α的含量降低(P<0.01),接近健康组。结果表明,优得乐对金黄色葡萄球菌性小鼠乳腺炎有明显的治疗作用。Staphylococcus aureus isolates were isolated from the milk of cow with clinical mastitis. Lactating mice were challenged by inoculation of the isolated S. aureus suspension via the teat canal. Ninety BALB/c mice were randomly divided into control group, model group and treatment group of thirty mice each. The mammary glands were harvested at indicated time points post-challenge with S. aureus. The pathology of mammary glands was histologically studied. Toluidine blue staining protocol was performed to investigate the distribution patterns of mast cells in mammary glands. The expressions of inducible nitric oxide synthase(iNOS) and 3-nitrotyrosine(3-NT) were detected by immunohistochernistry(IHC). Enzymelinked immunosorbent assay(ELISA) was conducted to measure the levels of tumor necrosis factor-α(TNF- α) in the mammary glands. Mouse mastitis model was developed at hour 24 after inoculation of S. aureus. It was clearly shown that neutrophils heavily occupied the dilated alveoli. The interlobular blood vessels were characteristic of congestion. The epithelial cells of some alveoli displayed markedly necrotic feature. In eontrast,neutrophil infiltration and congestion of some lobular vessels dramatically reduced in the treated mice. Compared with model mice,the number of mast cells decreased and mast cell degranulation was apparently inhibited. The expressions of iNOS and 3-NT were down-regulated. The levels of TNF-α were significantly reduced and nearly reached that in control mice. It was concluded that Youdele had a remarkable therapeutic effect on mastitis caused by S. aureus in mice.
