摘 要:目的检测在脂多糖(lipopolysaccharide,LPS)作用下,A549细胞中SP-B、NFкB、RARα表达情况,探讨感染因素(脂多糖)对SP-B表达及其有关调节因素(RARα)表达的影响,并观察在此过程中是否有NFкB表达的改变。方法培养的A549细胞分为对照组和实验组,实验组分别用不同浓度的脂多糖处理24h。免疫细胞化学法检测SP-B和RARα在各组A549细胞中的表达情况,免疫荧光共聚焦法检测NFкB在各组A549细胞中的表达变化。结果各实验组SP-B的表达均较对照组减弱(P<0.01),且呈药物浓度依赖性(P<0.05)。各实验组胞核内RARα表达均较对照组减弱(P<0.01),且呈明显的药物浓度依赖性(P<0.01)。而各实验组胞质RARα表达均较对照组增强(P<0.01),也呈药物浓度依赖性(P<0.01)。与对照组相比,实验组中NFкB部分转位至A549细胞胞核中表达。结论脂多糖作用后,A549细胞中SP-B的表达减少,且NFκB和RARα在细胞内的分布都发生了变化,表明在呼吸窘迫综合征的发病中,炎症过程可能起到重要作用,NFκB和RARα可能在此阶段中都参与了SP-B表达调控。Purpose To detect the expression and distribution changes of the RARα, NFκB and SP-B in A549 cells under LPS treatment. Methods Cultured A549 cells were divided into control and experimental groups. A549 cells were incubated with lipopolysaceharide (LPS) at different concentrations during 24 hours ,the expression of RARer and SP-B in all groups could be detected by immunocytochemical method, and the expression of NFκB in all groups could be detected by immunofluorescence copolymerization. Results Compared with control group, the expression of SP-B in cytoplasm of A549 cells was significantly decreased in the three experimental groups ( P 〈 0.01 ). LPS at different concentrations induced the expression of SP-B in A549 cells in a dose-dependent manner ( P 〈 0. 05 ). Compared with control group, the expression of RARα in nuclei was significantly decreased in the three experimental groups ( P 〈0. 01 ). LPS at different concentrations induced the expression of RARα in nuclei of A549 cells in a dose-dependent manner (P 〈 0. 01 ). Compared with control group, the expression of RARα in cytoplasm was significantly increased in three experimental groups (P 〈0. 01 ). LPS at different concentrations induced the expression of RARα in cytoplasm of A549 cells in a dose-dependent manner (P 〈0. 01 ). In control group, the expression of NFκB located in the cytoplasm of A549 cells. In experimental group, the partial expression of NFκB transferred to the nuclei of A549 cells. Conclusion The expression of SP-B decreases with the interference of lipopolysaccharide (LPS), and the distribution of NFκB and RARer has been changed in A549 cells. Thus, the inflammation possibly plays an important role in the patbogenesis of respiratory distress syndrome. In this stage, both NFκB and RARer regulate the expression of SP- B.
