摘 要:采用RT-PCR及RACE的方法从东方山羊豆中克隆了一种新的Na+/H+逆向转运蛋白基因。该基因全长2280bp,开放阅读框为1626bp,编码542个氨基酸残基。预测该蛋白分子量为60.0kDa,可能有10个跨膜区域。RT-PCR检测结果表明,在NaCl胁迫条件下东方山羊豆GoNHX1基因可在根、茎、叶中诱导表达,在叶中的表达量随着处理时间的延长而逐渐增加,到24h表达量下降。The full length cDNA of a new vacuolar Na^+/H^+ antiporter gene (GoNHX1) in Galega orientalis was cloned by RT-PCR and RACE. The data showed that the full length of GoNHX1 gene was 2 280 bp, the open reading frame of GoNHX1 was 1 626 bp and encoded a putative protein of 542 amino acid residues. The analysis showed that the putative protein was 60.0 kDa and had 10 potential transmembrane domains. RT-PCR results revealed that GoNHX1 expression could be induced in the roots, stems and leaves by NaC1. The expression level of GoNHX1 in leaves increased with the increasing of treatment time, but decreased after 24 h with the treatment of NaC1.
