摘 要:目的:探讨microRNAs(miRNAs)对原始卵泡的形成和发育的影响。方法:取1d龄新生小鼠卵巢,制备卵巢切片,利用原位杂交技术观察mir-143、let-7a、let-7b、let-7c和mir-125b在新生小鼠卵巢中的表达情况。结果:1d龄新生小鼠卵巢内只含有大量原始卵泡,mir-143、let-7a、let-7b、let-7c和mir-125b表达于这些原始卵泡的颗粒细胞内,在卵母细胞中不表达。结论:miRNAs可能通过调控原始卵泡颗粒细胞的增殖和/或分化,或通过调控下游靶基因表达直接或间接地影响卵泡形成和发育中的重要因子(FSH、抗苗勒氏管激素、生长分化因子9等),从而对原始卵泡的形成和生长启动产生影响。Objective: To explore the effect of micro RNAs (miRNAs) in the process during primordial follicular formation and subsequent follicular development in neonatal 1 d mouse ovary. Methods: Ovaries were collected from the postnatal mice on day 1 (P1) after birth. The expression of mir-143, let-7a, let-7b, let-7c and mir-125b was examined by in situ hybridization. Results: There were a large number of primordial follicles in neonatal mouse ovary. The miRNAs, mir-143, let-7a, let-7b, let-7c and mir-125b were detected in granulosa cells in ovary at P1. In contrast, these miRNAs were not detected in oocytes. Conclusion: These miRNAs in granulosa cells may be involved in some aspect of physiology that is unique or common for these granulosa cells, for example, cell proliferation and/or differentiation. In addition, these detected miRNAs may function as transcriptional regulator through their interaction with some endocrine, paracrine and/or autocrine factors such as FSH, AMH and GDF-9 which are important factors in micro-environment during folliculogenesis. These miRNAs are likely to paly a role in primordial follicle formation and growth.
