摘  要:以多糖得率为指标,选用正交试验对紫茎泽兰粗多糖的提取工艺进行优化,再经乙醇沉淀、脱蛋白和真空干燥后得到紫茎泽兰多糖(EAP)。结果表明紫茎泽兰粗多糖最佳提取工艺为:温度为100℃,在料液比1:20下提取2 h,提取2次。用脱氧核糖体系测EAP对·OH的清除作用;用硫代巴比妥酸法测小鼠肝组织丙二醛;用分光光度法测EAP对邻苯三酚自氧化体系的抑制作用。结果表明:EAP具有抗氧化活性,以抑制体外脂质过氧化效果最为显著。Optimization of extraction process of crude Eupatoriurn adenophorurn Spreng polysaccharides was investigated by orthogonal tests. Then the Eupatorium adenophorurn Spreng polysaccharide(EAP) was obtained after ethanol-precipitation, deproteinixation, and vacuum drying. The results showed that the optimum conditions for the isolation was as follows : times of extraction 2, time 2h, temperature 100°C, material to water ratio 1 : 20. The scavenging effect of EAP on hydroxyl radical ( · OH) was studied in deoxyribose system. The content of MDA in mice liver was measured by TBA assay. Self-oxidation inhibition was studied using the 1,2,3-phentriol while detected by spectrophotometric methods. The result showed that EAP had the effect of antioxidation, especially in markedly inhibition of the production of lipid peroxides in normal liver homogenate of mice.
