摘 要:目的探讨镉、镍单独及联合作用的发育毒性特点。方法应用小鼠胚胎肢芽细胞微团培养法观察CdCl2、NiCl2单独及联合染毒对胚胎肢芽细胞增殖和分化的影响。结果0.1~1.2μg/mlCdCl2或5.5~27.5μg/mlNiCl2对胚胎肢芽细胞的增殖均有明显的抑制作用;0.4~1.2μg/mlCdCl2或5.5~27.5μg/mlNiCl2也可明显抑制胚胎肢芽细胞的分化,但CdCl2在0.1和0.2μg/ml时可促进细胞分化。CdCl2和NiCl2联合染毒对胚胎肢芽细胞的增殖和分化的抑制分别表现为相加和加强作用。结论Cd和Ni可能通过某种共同机制而对胚胎肢芽细胞的增殖和分化的抑制产生协同作用。To investigate the single and combined developmental toxicity of cadmium and nickel,mouse limb bud cell micromass culture was conducted to observe the effects of CdCl2 and NiCl2 on the proliferation and differentiation of mouse embryo limb bud cell.The results showed that CdCl2 at dosage of 0.1 ̄1.2 μg/ml and NiCl2 at dosage of 5.5 ̄27.5 μg/ml significantly inhibited the increase of cell number,CdCl2 at dosage of 0.4 ̄1.2 μg/ml and NiCl2 at dosage of 5.5 ̄27.5 μg/ml also reduced the number of cartilaginous foci,however,CdCl2 at dosage of 0.1 μg/ml and 0.2 μg/ml could increase the number of cartilaginous foci.These two metal ions in combination could affect the proliferation and differentiation of mouse embryo limb bud cell by adding together their inhibitory effects and potentiating the inhibition respectively.It is suggested that Cd and Ni in combination exhibited their effects on the inhibition of proliferation and differentiation of mouse embryo limb bud cell in a synergistic way.
