摘 要:[目的]系统研究脱水法合成6-苄氨基嘌呤(6-BA)的反应条件。[方法]以一定比例的腺嘌呤和苯甲醇为原料制备6-BA,用紫外分光光度计测定产品含量。[结果]随着反应时间的增加,反应物中产品的含量逐渐增大,2.5h以后,产品含量增加速度变缓,3.0h以后基本不再增加。PEG的用量在0.5%时,产品产率最高。当氢氧化钠与腺嘌呤的摩尔比小于1.0:1.0时,得不到产品。当氢氧化钠与腺嘌呤的摩尔比大于1.0:1.0时,产率很高,并且氢氧化钠的用量与产率无明显的相关关系。当氢氧化钠与腺嘌呤的摩尔比大于1.1:1.0时,产率基本不变。[结论]脱水法合成6-苄氨基嘌呤的最佳反应条件为:腺嘌呤:NaOH:苯甲醇:PEG400=1.0:1.1:6~7:0.03,反应时间为2.5~3.0h,反应温度低于250℃。[ Objective ] The purpose was to research the reaction condition of synthesizing 6-benzylaminopurine (6-BA) by dehydration method systematically. [ Method] The 6-BA was prepared with adenine and benzyl alcohol at a certain proprotion as raw materials and the content of product was determined by UV-specrophotometer. [ Result] As the reaction time was increased, the content of product in reactants was increased gradually. After 2.5 h, the in- creasing speed of product content became slower. After 3.0 h, the content of product was no longer increased basically. When the dosage of PEG was 0.5%, the yield of product was highest. When the molar ratio of sodium hydroxide to adenine was smaller than 1.0:1.0, the product could not be got. When the molar ratio of sodium hydroxide to adenine was bigger than 1.0:1.0,the yield was very high and there was no obvious correlation between the dosage of sodium hydroxide and yield. When the molar ratio of sodium hydroxide to adenine was bigger than 1.1 : 1.0, the yield was basically invariable. [Conclusion] The optimum reaction condition of synthesizing 6-BA by dehydration method was as follows: the ratio of adenine, sodium hydroxide, bertzyl alcohol and PEC400 was 1.0:1.1:6- 7:0.03, the reaction time was 2.5 - 3.0 h and the reaction temperature was lower than 250 ℃.
