摘 要:目的探讨丹龙醒脑片对大鼠脑缺血再灌注海马区细胞凋亡指数(AI)、Fas/FasL、TNF-α表达的影响。方法运用大脑中动脉栓塞再通法建立脑缺血再灌注模型,将动物随机分为假手术组、模型组、丹龙醒脑片组、尼莫地平组,采用原位末端标记法(TUNEL)检测凋亡神经细胞。采用免疫组织化学法,观察海马区Fas/FasL、TNF-α的表达。结果丹龙醒脑片能显著减轻神经细胞的损伤;假手术组有一定量的Fas/FasL、TNF-α表达,缺血后Fas/FasL、TNF-α迅速增加,丹龙醒脑片组显著抑制Fas/FasL、TNF-α的表达,模型组和丹龙醒脑片组、尼莫地平组比较,差异均具有统计学意义(P<0.05)。结论抑制由缺血再灌注损伤诱导的Fas/FasL、TNF-α表达可能是丹龙醒脑片具有脑保护作用的机制之一。Objective To investigate the effect of Danlong Xingnao tablet (DLXNT) on expression of AI, Fas/FasL and TNF-α after cerebral ischemic-reperfusion in hippocampus in rats. Methods The model of cerebral ischemia-reperfusion was induced by middle cerebral artery occlusion (MCAO) and recanalization. The rats were randomly divided into four groups including sham-operation, control, DLXNT and nimodipine groups. The expression of AI, Fas/FasL, TNF-α in hippocampus were detected with TUNEL and immunohistochemistry. Results DLXNT could reduce obviously the injury of nervous cell; expression of Fas/FasL and TNF-α positive cells existed in sham-operation group, Fas/FasL and TNF-α were increased immediately after cerebral ischemic-reperfusion; compared with model group, the expressions of Fas/FasL and TNF-α were remarkably inhibited in DLXNT group(P〈0.01, P〈0.05). Conclusion The effect of DLXNT maybe inhibiting the expression of Fas/FasL and TNF-α induced by cerebral isehemia-reperfusion injury.
