摘  要:目的探讨邻苯二甲酸二乙基己基酯(DEHP)对大鼠肝细胞和睾丸细胞DNA的损伤。方法选用雄性Wis-ter大鼠25只,随机分为5组,每组5只,通过灌胃的方法摄入DEHP,4个染毒组剂量浓度分别为1500、3000、4500、6000mg·kg-1.d-1,1个阴性溶剂对照组,用单细胞凝胶电泳技术(彗星试验)检测大鼠肝细胞和睾丸细胞DNA的损伤。结果各染毒组和对照组大鼠肝细胞DNA损伤率分别为28.0%、69.0%、86.0%、94.0%和18.0%,各染毒组肝细胞DNA损伤程度与对照组比较差异有显著性(P<0.05,P<0.01),且大鼠肝细胞损伤率与染毒剂量之间呈剂量-反应关系(r=0.942,P<0.01);各染毒组和对照组大鼠睾丸细胞DNA损伤率分别为29.0%、71.0%、89.0%、95.0%和15.0%,各染毒组睾丸细胞DNA损伤程度与对照组比较差异有显著性(P<0.05,P<0.01),且大鼠睾丸细胞损伤率与染毒剂量之间呈剂量-反应关系(r=0.930,P<0.01)。结论DEHP可造成大鼠的肝细胞和睾丸细胞DNA的损伤,并随染毒剂量的增加有加重的趋势。Objective To study the effect of di-(2-ethylhexyl) phthalate (DEHP) on DNA damage in liver and testicle cells of rats. Methods Wister male rats were divided into 5 groups,including 4 experimental group and a control group. The experimental groups were exposed at the doses of 1 500,3 000,4 500 and 6 000 mg·kg^-1 ·d^-1 DEHP. The degree of DNA damage in the liver and testicle cells were detected by single cell gel electrophoresis. Results The rates of DNA damage in liver cells of each experimental group and the control group were 28.0% ,69. 0% ,86.0% ,94. 0% ,and 18.0% ,respectively,with a significantly higher rates of DNA damage observed in the experimental groups ( P 〈 0. 05, P 〈 0.01 ) and a significant dose-response relation between the rates of DNA damage and the doses of DEHP( r = 0. 942 ,P 〈 0. 01 ). The rates of DNA damage in testicle cells of each experimental group and the control group were 29. 0% ,71.0% ,89.0% ,95.0% ,and 15.0% ,respectively, showing the rates of DNA damage in each experimental group were significantly higher than that in the control group( P 〈 0. 05, P 〈 0. 01 ), and showing a significant dose-response relation between the rates of DNA damage of testicle cells and the doses of DEHP( r = 0. 930, P 〈 0.01 ). Conclusion DEHP could cause DNA damage in liver and testicle cells of rats and the degree of DNA damage was aggravated with the increase of DEHP.
