摘 要:采用密闭式脐血采血法收集了192份脐血,对脐血的采集量、有核细胞数、经淋巴细胞分离液分离后的单个核细胞的回收率进行了评估用含有多种生长因子的半固体培养基对脐血所含的粒单祖细胞及红系祖细胞进行了检测,并和骨髓的同类祖细胞作了比较.结果,脐血的平均可采集量为99mL,每份脐血所含有的有核细胞数为(136±48)×108,分离后单个核细胞的回收率为(423±164)%,脐血的粒单祖细胞及红系祖细胞的集落产率均高于骨髓,差异有显著性,(P<005).脐血形成的粒单及红系集落体积也较骨髓的同类集落体积大.Airtight-collecting method was applied in the collection of 192 umbilical cord blood samples, in which evaluated were the volume, nucleated cell number, recovery rate of mono-nucleated cells after Ficoll separation. With semisolid culture containing growth factors, granular monocyte and erythroid lineage progenitor from cord blood were tested and analyzed with those from bone marrow. The results were indicated as follows: average collection volume was 99 mL; nucleated cell number was 13.6_4.8*108 for each blood sample, recovery rate of mono-nucleated cells was (42.3±16.4%) after Ficoll separation; colony formation rate of granular monocyte and erythroid lineage progenitor was higher in umbilical cord blood than in bone marrow, with significant difference (P<0.05); and colony formation size of granular monocyte and erythroid lineage progenitor was higher in umbilical cord blood than in bone marrow, with significant difference (P<0.05). All the above suggested that umbilical cord blood contains more hemopoietic progenitors than bone marrow.
