摘 要:为了从成熟的肥城桃果实中提取纯净完整的RNA,本试验在原CTAB法的基础上进行了改进,并对RNA的完整性、产率和纯度等方面进行评价。结果表明,采用改良CTAB法所得RNA完整性好,条带清晰,无明显降解,28S和18S rRNA的亮度比约为2∶1,A260/A280达1.89,且产率较高,为356.21μg/g,经RT-PCR后得到一特异条带,表明该RNA可用于后续分子生物学操作。In order to extract pure and integrate RNA from Feicheng peach fruit, an improved CTAB method was developed, and the integrity, productivity and purity of the extracted RNA were evaluated in this paper. The results showed that the RNA extracted by the improved CTAB method had good integrity and the RNA bands were clear without degradation. The ratio of the brightness of 28S rRNA to that of 18S rRNA was approximately 2:1 ; the A260/A280 was 1.89, and the yield of total RNA was 356.21ug/g. After RT - PCR, a specific band was obtained, indicating that the extracted RNA could be used for the following molecular biology operations.
