摘 要:目的观察1周力竭游泳运动后大鼠下丘脑室旁核(PVN)NF-κB基因的表达变化及变化规律。方法将35只大鼠随机分为对照组(n=5)和力竭组(n=30),力竭组大鼠进行1周力竭游泳运动,最后一次力竭运动结束后分即刻(0h),6h,10h,14h,20h,24h共6个时间点,采用组织化学SABC法观察大鼠下丘脑室旁核内NF-κB核转录因子的定位和分布表达规律,采用Western Blot法研究下丘脑室旁核NF-κB含量变化,通过图像分析系统和统计学软件进行图像和数据分析。结果①Western Blot显示对照组大鼠下丘脑室旁核NF-κB表达极低,而运动结束后6h,10h,14h NF-κB表达明显升高,10h时达到最大值;②SABC法显示对照组大鼠下丘脑室旁核NF-κB阳性神经元呈少量散在分布;与对照组相比,力竭组室旁核NF-κB表达在6h开始升高(P<0.05),10h NF-κB阳性神经元数目显著升高达峰值(P<0.05),14-20h开始明显回落,仍高于对照组(P<0.05),24h恢复到正常水平(P>0.05)。结论力竭游泳运动后,大鼠下丘脑室旁核NF-κB的表达增多,说明NF-κB可能参与机体中枢神经系统对运动的调节。Objective To observe the expression pattern of nuclear factor-kappa B (NF-κB)in paraventricular nucleus (PVN)of hypothalamus in rats after a week exhausted swimming exercise. Methods Thirty-five rats were randomized into control group(n = 5)and exhausted swimming group(n=30). The model rats were established by a week exhausted swimming exercise. Expression of NF-κB PVN was detected by immunohistochemical SABC and Western Blot at 0 h,6 h, 10 h, 14 h,20 h,24 h after a week exhausted swimming exercise, and analyzed by imaging analysis system and statistic software. Results ①The expression of NF-κB was extremely low in PVN in controls, but increased significantly after exhausted swimming exercise ,and peaked at 10 h after exhausted swimming exercise. ②Few NF-κB in control group dispersively distributed in PVN. Compared with control group, the numbers of NF-κB in PVN increased significantly at 6 h after exhausted swimming exercise ( P 〈 0.05 ) , peaked at 10 h, and then decreased at 14 - 20 h ( P 〈 0.05 ). The numbers of NF-κB descended to the normal level at 24 h(P 〉 0. 05). Conclusion After exhausted swimming exercise, the expression of NF-κB increases in PVN of hypothalamus in rats ,which demonstrates that NF-κB may participate in the regulation of movement in CNS.
