摘 要:目的探讨氧化苦参碱对大鼠肾间质纤维化的影响。方法将48只SD雄性大鼠随机分为假手术(SOR)组、单侧输尿管梗阻(UUO)组、血管紧张素Ⅱ受体拮抗剂氯沙坦组(LOS组)和氧化苦参碱组(OM组)。大鼠肾组织行苏木精-伊红和Masson染色,计算肾间质纤维化的相对面积。免疫组化技术检测ColⅠ和FN在肾组织中的表达。结果OM治疗组和LOS治疗组能有效改善UUO大鼠的肾脏间质纤维化,OM组和LOS组中各时间点ColⅠ和FN蛋白的表达均分别低于UUO组(P<0.05),但高于SOR组(P<0.05),两组治疗间差异无统计学意义(P>0.05)。结论氧化苦参碱在一定程度上可减轻UUO大鼠肾间质纤维化程度,延缓肾脏间质纤维化。Objective To observe the effect of oxymatrine on renal interstitial in unilateral ureteral obstruction(UUO)rat. Methods 48 Sprague-Dawley male rats were randomly divided into sham-operated group( SOR group), unilateral ureteral obstruction model group(UUO group), losartan group(LOS group) and oxymatrine group(OM group). The area of renal interstitial fibrosis was calculated by HE and Masson staining,while the expression levels of FN and Col Ⅰ were detected by immunohistochemistry. Results Renal interstitial fibrosis was improved significantly in both LOS group and OM group. When compared with UUO group, the expression levels of Col Ⅰ and FN were reduced in the 2 groups, which were the lowest in SOR group(all P 〈 0.05. However, there was no diffference observed between LOS and OM group. Conclusion Oxymatrine may reduce the damage and delay the progress of renal interstitial fibrosis in UUO rat model.
