摘  要:目的通过腹腔注射荷包牡丹碱(BIC)阻断GABA受体,观察海马CA1区神经元超微结构、氧化还原状态和下丘脑GABA的变化,探讨运动过程中HPA轴的中枢调节机制,并进一步探索这些指标在运动疲劳产生机制中的作用。方法腹腔注射BIC或生理盐水的SD大鼠进行一次性力竭游泳,并分别在运动前、运动后即刻1、h、3 h和24 h断头取材,用放免法测定血清C,DNS-Cl荧光光度法测定下丘脑GABA含量,顺磁共振波谱法测定PBN自旋捕获剂捕获的海马CA1自由基含量,透射电镜观察神经元超微结构。结果(1)BIC组大鼠平均力竭运动时间较对照组明显缩短(P<0.05)。(2)BIC组各时间点海马CA1区.OH的产生都较对照组的相应值高,运动后1 h和3 h时差异显著(P<0.05)。(3)两组大鼠的下丘脑GABA含量变化趋势类似。(4)BIC组血清C浓度值在运动后即刻明显较对照组相应值高(P<0.05),运动后3 h显著低于对照组的相应值(P<0.05)。结论(1)腹腔注射BIC可阻断大鼠海马GABA-A受体,此作用具有时段延续性和可逆性。(2)GABA通过GABA-A受体抑制运动应激时自由基过量生成对大鼠海马CA1神经元超微结构产生保护作用。(3)GABA的大量增加不是引起运动能力下降直至力竭的直接原因。To determine the potential role of hippocampus and hypothalamus in the center regulating mechanism of neuroendocrine when sports fatigue, rats were injected with bicuculline one kind of blockers of GABA receptors, and fluorescence analysis method was used to detect hypothalamus GABA concentration, electron paramagnetie resonance (EPR) and transmitted electron microscope ware used to the observe free redical generation and the morphological changes in hippocampus CA1 neurons respectively, simultaneously radioimmunity was applied to analysis the activity of HPA axis in recovery phase after acute exhausting swimming. The results showed that ( 1 )The average swimming time of rats in BIC groups was much shorter than controls. (2)Free radical Generation in hippocampal CAI of BIC rats was more than corresponding values of control groups. (3)The changes of GABA concentration in h.vpothalamus were similar between the two big groups. (4)The level serum C induced by exhausting swimming after BIC injection was higher than the level of control groups. The results of this experiment suggested that ( 1 ) Peritonal injection of BIC was able to block GABA - A receptors in hippocampus of rats, and the effect was persistent and reversible. (2) Increase of free radical generation induced by exhausting exercise was one of the principal causes to hippocampus CA1 neurons morphological changes; GABA could suppress the production of free radical induced by exercise stress, and play the role of protection for center neurons. (3)Over generation of GABA was not the direct cause for exhaustion.
