摘  要:为了研究大气细颗粒物(PM2.5)对雄性大鼠的生殖毒性作用,将不同剂量的PM2.5颗粒物悬液采用气管滴注方式对雄性Wistar大鼠分组染毒,染毒剂量分别为0、1.6、8.0、40.0mg·kg-(1BW),24h后应用流式细胞术对睾丸组织细胞周期及DNA倍体进行检测分析.结果显示:PM2.5对大鼠睾丸组织各倍体细胞比例和细胞周期有明显影响,可导致二倍体细胞数显著降低(p<0.05);可引起以二倍体细胞为主的G0/G1期细胞比例显著下降(p<0.05),G2/M期细胞比例和细胞增殖指数(PI)显著上升(p<0.05).结果提示,PM2.5可透过血睾屏障,干扰细胞周期进程,对生殖系统具有一定的毒性作用.In order to study the effects of PM2.5 on testis cells of male rats and explore the toxic mechanism of PM2.5 on the genital system, male Wistar rats (SPF Grade )were exposed to PM2.s by intratracheal instillation at various doses (0, 1.6、8.0、40.0mg·kg^-1(BW)). After 24h exposure, the cell cycle and the DNA ploidy were examined by Flow cytometry (FCM). Results showed that PM2.5 had significant effects on the cell percentage and the cell cycle of testis spermatogenic cells in rats. The percentage of diploid cells and the cell number of G0/G1 phase decreased significantly(p〈0.05), and the cells of G2/M and cell proliferation index(PI)increased significantly(p〈0.05 )compared with the control after PM2.5 exposure. These results indicated that PM2.5 could permeate blood-testis barrier into germ cells of male rats and induce genital system injury by affecting the proceeding of cell cycle.
