甲状腺激素对仔鼠胸腺上皮细胞的影响

摘  要:【目的】研究给予甲状腺激素后,仔鼠胸腺上皮细胞的变化。【方法】Wistar母鼠在发现阴栓后随机分为2组:对照组和实验组。各组均在发现阴栓后15d灌胃,直到仔鼠生后20d。实验组采用浓度为50g/L的甲状腺素,对照组每天给以等量生理盐水。两组仔鼠分别在生后10、20、40d处死,取胸腺,测定仔鼠的胸腺指数,血清FT3、FT4水平,免疫组化染色显示CK10&13阳性胸腺上皮细胞的变化。【结果】与对照组相比,实验组10d龄仔鼠的胸腺指数降低,FT3显著升高,交界处和髓质中胸腺上皮细胞的数密度降低,被膜下上皮细胞的平均灰度增高,交界处细胞的平均灰度降低;20d龄仔鼠交界处上皮细胞的数密度降低,皮质上皮细胞的平均灰度增高;40d龄仔鼠的交界处上皮细胞的数密度降低,平均灰度增高,其余指标未见明显异常。【结论】胸腺上皮细胞存在异质性,不同部位的胸腺上皮细胞对甲状腺激素的反应各不相同,尤其是以交界处和髓质的上皮细胞变化最大。[Objective] To detect the changes of thymic epithelial cells (TEC) in infant rats after administering mother rats with thyroid hormones. [Methods] Wistar mrs were divided randomly into normal group and experimental group after finding vaginal Hug, rats were administered by gavage from with 50g/L tetraiodothyroine the 15th gestational days to 20th days after birth, for experimental Pregnant rats and Physiologicalsaline for normal grouP. Infant rats were sacrificed respectively on 10th respectively , 20th , 40th days respectively. Thymic index and thyroid hormones FT3 , FT4 were measured, Immunohistochemical studies on TEC subsets defined by anticytokeratin 10&13 monoclonal antibody. [ Results] The thymie index decrease and FT3 level increase in experimental group on 10th days. Numerical density on area (NA) of TEC were lower at cortieo - medullary junction on 10th , 20th and 40th days and also at medulla on 10th days, Average grey density of TEC increase at s^sualr TEC on lOth days , cortical TEC on 20th days, eortieo-medullary junction on 40th days, decrease at cortico- medullary junction on 10th days. [ Conclusions] The result demonstmtesthat the TEC heterogeneity, TEC in different and have various reactions to thyroid hormones, especially TEC at cortieo-medullary junction and medulla change greatly.
