摘  要:【目的】采用高效液相色谱法建立和改良测定人血清中胺碘酮的方法学。【方法】色谱柱:shim-packvp-ODS柱(150mm×4.6mm,5μm)。流动相为甲醇∶乙腈∶乙酸铵缓冲液=45∶45∶10(v/v/v),流速1.0ml/min,检测波长242nm。最低检出限为0.125μg/ml。【结果】纯甲醇色谱峰出现在进样后1.809min,AD标准品色谱峰出现在进样后10.421min。以峰面积与标准浓度做回归,得回归方程Y=2.67049×10-6X-0.1619,r=0.962(n=10)。线性范围为:0.125~4μg/ml。日内变异系数为2.1%~5.1%,日间变异系数为3.2%~7.9%。【结论】此法是较为简便可行的检测血清胺碘酮浓度的方法。[ Objective] To establish A HPLC method for improve the determination of amiodarone in human serum. [Methods] The column was shim-packvp-ODS (150 mm x 4.6 mm, 5 μm) . The mobile phase was methanol-acetonitrile-acetamide (45:45 :10, v/v/v), pH value was adjusted to 5.0 by acetic acid. The detection wavelength was 242 nm and. LOD was 0.125 μg/ml. [Results] HPLC peak of pure methanol appeared at 1.809 min after sample injection, and peak of AD at 10.421 min. Regression equation of peak areas and concentration was noted as Y = 2.67049 × 10^-6X- 0.1619, r = 0.962 ( n = 10) . The intraday and interday CV was 2.1% - 5.1% and 3.2 %- 7.9 % respectively. [ Conclusions] This chromatography condition is practicable in clinic monitoring the amiodarone concentration.
