摘 要:目的探讨促红细胞生成素(EPO)对2型糖尿病大鼠肾损伤的保护作用。方法4周龄雄性SD大鼠60只随机分为2组。正常对照组10只,普通饲料喂养;糖尿病模型组50只,单侧肾切除手术2周后,高脂饲料喂养,喂养4周后尾静脉注射小剂量链脲佐菌素(STZ)35mg/kg,注射STZ30d后,将其随机分为实验组和治疗组,治疗组给予促红细胞生成素300U/kg腹腔注射。30d后,测量24h尿蛋白含量,并用免疫组化染色观察肾脏CD34的阳性表达。结果实验组及治疗组24h尿蛋白含量及肾脏CD34的表达均高于对照组(P<0.05),治疗组24h尿蛋白含量及肾脏CD34的表达均低于实验组(P<0.05)。结论EPO对2型糖尿病大鼠的肾损伤具有保护作用。Objective To study of the effects of Erythropoietin on the rat model of type 2 Diabetic Nephropathy. Methods 60 male SD rats with 4 -week old were randomized into two groups: 10 in the control group (CG) feeding with norreal forage, 50, two weeks after the unilateral nephrectomy, in the DM group feeding with high grease forage. Intravenous injection of STZ after feeding 4 weeks (35mg/kg), after 30 days divide it to two groups randomly, experimental group (EG) and treatment group (TG). TG rats were given EPO 300U/kg peritoneal injection. All the rats were tested with the 24h urine protein and immunohistochemistry staining was adopted to observe the positive expression of renal CD34 molecular. Results The 24h urine protein and the positive expression of renal CD34 molecular with EG and rig were significantly higher than that in the control group ( P 〈 0. 05), and TG was lower than EG ( P 〈 0. 05). Conclusion EPO has the protection on the rat model of type 2 Diabetic Nephropathy.
