不同年龄大鼠胃肠道G细胞和ACTH-IR细胞的变化

摘　　要：目的：了解大鼠胃肠道胃泌素（ｇａｓｔｒｉｎ，Ｇ）细胞和促肾上腺皮质激素免疫反应性（ａｄｒｅｎｏｃｏｒｔｉｃｏｔｒｏｐｈｉｃｈｏｒｍｏｎｅｉｍｍｕｎｏｒｅａｃｔｉｖｉｔｙ，ＡＣＴＨＴＲ）细胞年龄变化特点及胃肠道与脑垂体ＡＣＴＨＩＲ细胞年龄变化的关系。方法：应用ＰＡＰ免疫组织化学方法和胶金免疫电镜技术。结果：１８个月大鼠胃窦Ｇ细胞及十二指肠和脑垂体ＡＣＴＨＩＲ细胞数量均分别显著低于其它组（Ｐ＜００５；Ｐ＜００１）。２５个月大鼠十二指肠ＡＣＴＨＩＲ和胃窦Ｇ细胞数量为最高值，脑垂体ＡＣＴＨＩＲ细胞数量最高值为３５天大鼠。此外，在胶金免疫电镜下，可见十二指肠杯状细胞内含有胶金颗粒。结论：大鼠十二指肠和脑垂体ＡＣＴＨＩＲ细胞及胃窦Ｇ细胞数量均有明显的年龄变化；胃肠道Ｇ细胞与ＡＣＴＨＩＲ细胞数量随年龄变化是一致的，而胃肠道与脑垂体ＡＣＴＨＩＲ细胞数量与年龄变化却不相一致；胃肠道的一些杯状细胞可能有分泌ＡＣＴＨＩＲ物质的功能。Objective: To observe changes of gastrin (G) cell and adrenocorticotrophic hormone immunoreactivity (ACTH-IR) cell in the gastrointestinal tract with age and the relationship of age chamges between the gastrointestinal and pituitary ACTH-IR cells of the rat.Methods:With the PAP immunohistochemical method and colloidal gold immunoelectron microscopy.Results: The numbers of G cells in gastric antrum and ACTH-IR cells in the duodenum and pituitary were markedly lower in eighteen months group than that in other groups (P<0.05;P<0.01),respectively.The numbers of duodenal ACTH-IR cells and antral G cells were the highest in 2.5 months group,but the numbers of pituitary ACTH-IR cells were the highest in 35 days group.In addition,the gold particles were seen in goblet cells of the duodenum by colloidal gold immunoelectron microscopy.Conclusion:Changes of the numbers in duodenal and pituitary ACTH-IR cells and antral G cells of the rat all were marked with age;Age changes in the numbers between G cells and ACTH-IR cells in gastrointestinal tract were the same,that of ACTH-IR cells between the gastrointestinal tract and the pituitary were different;A number of goblet cells in the gastrointestiaal tract may have ACTH-IR substance secreting function.
