摘 要:目的从脑内氨基酸类递质角度探讨康痫丸治疗原发性癫痫的作用机制。方法将40只SD大鼠随机分为正常对照组、癫痫模型组、丙戊酸钠组、康痫丸组,每组10只。用戊四氮(PTZ)亚惊厥剂量造模。通过免疫组化观察药物处理后致痫大鼠海马CA1、CA3、齿状回和顶叶大脑皮层的γ-氨基丁酸(GABA)、谷氨酸(Glu)表达强度。结果PTZ致痫后,大鼠海马及皮层GABA阳性细胞数及光密度减少,而Glu阳性细胞数及光密度增加,提示PTZ同时影响脑内GABA和Glu的代谢,证实了GABA及Glu与癫痫的相互关系。采用康痫丸抗癫痫,动物海马、皮层GABA阳性细胞数及光密度增加,而Glu阳性细胞数及光密度减少。结论康痫丸降低脑内Glu,升高GABA水平可能是其发挥抗癫痫作用的机制之一。Objective: To study the mechanism of Kangxian Pill in the treatment of epilepsy by the amino - acid neurotransmitter in the brain tissue. Methods: 40 SD rats were randomly divided into 4 groups: normal group, model group, sodium valproate group and Kangxian Pill group. The pentylenetetrazol (PTZ) was used to establish epileptic model. The GABA and Glu of epileptic rats' brain were examined. Results: After PTZ induced, the GABA positive cell count and optical density decreased while the Glu positive cells and optical density increased in rats' hippocampus and cortex. It suggested that the PTZ impacted the brain' s metabolism of GABA and Glu at same time. The relationship of GABA and Glu between epilepsy confirmed. After Kangxian Pill treated, the GABA positive cell count and optical density increased while the Glu positive cells and optical density decreased in rats' hippocampus and cortex. Conclusion: Kangxian Pill can increase GABA and decrease Glu in brain that might one of mechanisms of Kangxian Pill anti - epilepsy.
