摘 要:为研究肝素对胸腺细胞发育的影响,以FTOC的方法进行体外器官培养,以不同浓度肝素干预其发育。观察发育不同时间点,胸腺细胞数和胸腺细胞表型的变化情况。结果显示:在4U/ml的肝素浓度下,胸腺细胞数比对照组显著减少,尤其是CD4+CD8+双阳性细胞数变化明显,且胸腺细胞数的减少与肝素用量具有明显剂量依赖关系;另外,在体外培养的第3天,肝素干预组中胸腺细胞37.98%表达CD69分子,显著高于对照组的0.76%。这些证据提示肝素能够影响双阳性胸腺细胞的发育。To investigate the effect of heparin on thymocytes development, the fetal thymus organ culture(FTOC) with or without heparin, was performed and at various days of the culture, thymocytes were harvested to count and stained with monoclonal antibodies for phenotype analysis by FACS. The results showed that heparin dramatically reduced the thymocytes and exhibited a dose dependent curve. Meanwhile, the proportions of subpopulations in thymus, especially CD4^+ CD8^+ subpopulation dramatically decreased in the present of heparin. To explore the possible mechanism, expression of CD69, one of development markers for double positive thymocytes, was detected on day 3 in FTOC. Results showed a higher expression of CD69 on cultured thymocytes with heparin compared to that of without heparin, suggesting that heparin may interfere in the processing of double positive thymocytes development to single positive thymocytes. In one word, heparin interfered in the development of thymocytes by blocking the development from double positive thymocytes to single positive thymoeytes in the thy mus.
