摘  要:γδ T细胞和αβT细胞是两种表型和功能不同的T淋巴细胞亚群。为研究两种细胞在抗原特异性免疫应答中是否具有协同作用,并进一步探讨其相互作用的方式,本文利用实验性自身免疫性葡萄膜炎小鼠模型获得脾脏和淋巴结T细胞,用γδ TCR和αβTCR单克隆抗体染色,分别用带有磁珠的抗FITC和抗PE单克隆抗体通过阳性或阴性选择获得高度纯化的γδ T细胞和αβ T细胞,观察不同比例的γδT细胞和αβT细胞在体外共孵育后IRBP1-20抗原特异性增殖和细胞因子表达,并用细胞培养薄膜将二者分开,测定细胞因子表达情况。结果表明,γδ T细胞或αβT细胞(99%纯度)都不能单独发生抗原特异性免疫应答。只有在二者比例为5%γδT细胞和95%αβT细胞时能产生最大的抗原特异性反应。当两种细胞脱离接触时,不能出现抗原特异性反应。γδ T细胞或αβ T细胞的培养上清也不能使γδT细胞或αβT细胞发生抗原特异性反应。本研究提示,小鼠γδT细胞和αβT细胞在抗原特异性免疫应答中具有协同作用,这种协同作用是通过两种细胞相互接触,γδT细胞协助αβT细胞产生免疫应答实现的。To investigate the synergistic role of γδ T cells and αβ cells, the highly purified γδ T cells and αβ cells were obtained from spleen and lymph nodes from B6 mice induced for AEAU after cultivation in vitro and selected with MACS. Then, they were pooled together with various ratio from 99% to zero. Furthermore, they were separated by cell culture insert, so that they could not contact each other. During cultivation with specific antigen IRBP1-20 for 48 hours, the supernatants of culture were collected and the proliferation and cytokine production of the cultivated cells were measured. It was found that neither γδ T cells nor αβ cells in 99% purity could independently react with the specific antigens, except that the ratio of γδ T cells and αβ cells was 5 % : 95 %, during which the maximal antigen-specific response occurred. No antigen-specific reaction occurred, when these two T cell subsets lost contact each other. Also, the cultural supernatants of γδ T cells or αβ T cells were unable to induce the antigen-specific responses. These results suggest that γδ T cells and γδ T cells display synergistic interaction in immune responses to specific antigens through contacting each other hetween these two T cell subsets, in which γδ T cells may help αβ T cells in the responses against the specific antigens.
