摘  要:目的研究胆汁对体外培养条件下人成纤维细胞的作用,以探讨胆汁在胆道瘢痕形成过程中的作用及其机制。方法术中抽取胆囊息肉病人的胆汁,过滤除菌后稀释成不同浓度加入到体外培养的人成纤维细胞中,观察和检测胆汁对成纤维细胞形态学、细胞增殖活性、细胞周期及凋亡的影响。结果形态学观察显示胆汁促进成纤维细胞增殖和表型转化。MTT检测结果显示,成纤维细胞的光密度值随着加入胆汁浓度的升高而升高,且呈现出一定程度的浓度依赖效应。流式细胞仪检测结果显示,与对照组相比,实验组成纤维细胞凋亡率下降,G0/G1期细胞比例下降,S期细胞比例增加。结论胆汁促进胆道瘢痕的形成和胆道瘢痕性狭窄的产生。完整的胆道黏膜在防止胆汁对成纤维细胞作用方面起到了重要的屏障作用。Objective To study the effect of bile on the cultured human fibroblasts to explore the role of bile in the formation of hiliary scars and relative mechanisms. Methods Human bile was collected during the operations for patients with polypoid lesion of gallbladder. After being percolated, the bile of different concentrations was added into the culture medium. Morphological changes in both control group and trial group were observed under inverted microscope and electron microscope. The effect of different concentrations of bile on the proliferation of cultured fibroblasts was studied by MTT assay. The apoptosis rate and cell circle distribution were determined by FCM. Results Morphological observations revealed that the proliferation of fibroblasts in trial group was enhanced and the phenotype modulated by bile. The result of MTT assay showed that the correspondent OD values became larger when the concentrations of bile became higher. And the effect was concentration-dependant to some extent, FCM results showed that the apoptosis rates of cells and the proportion of cells in G0/G1 stage in trial group were lower and the proportion of cells in S stage in trial group was higher than in the control group. Conclusion Human bile enhances the formation of biliary scar and biliary stricture. The integrity of biliary mucous membrane barrier plays an important role in preventing the effect of bile on fibroblast cells.
