摘 要:目的:对急性子中2种主要凤仙萜四醇苷类成分凤仙萜四醇苷K、凤仙萜四醇苷A进行含量测定。方法:采用反相高效液相色谱-蒸发光散射检测法,使用Eclipse XDB-C18色谱柱(4.6mm×250mm,5μm),流动相为甲醇-水(梯度洗脱,0~22min时为50∶50,22~50min时为54∶46),流速0.8mL·min-1,柱温25℃。ELSD检测器条件:载气(空气)流速2.5L·min-1,漂移管温度89℃。结果:凤仙萜四醇苷K、A进样量在5~20μg范围内线性关系良好,相关系数(n=6)分别为0.9995和0.9992;平均回收率(n=6)均为98.0%,RSD均为1.3%。结论:本方法能够准确测定急性子中凤仙萜四醇苷K、A的含量,方法简便,结果可靠。Objective:To determine two hosenkosides( hosenkoside K and hosenkoside A)in the seeds of Impatiens balsamina. Methods:Using RP- HPLC with ELSD method. Separation was carried out on Eclipse XDB -C18 column(4.6 mm×250 mm,5 μm). A linear solvent gradient was used,with a mobile phase of methanol - water(0 - 22 rain ,50:50 ;22 -50 min ,54:46 ). The flow rate was 0.8 mL·min^-1 at 25℃. The gas(air) flow rate was 2. 5 L·min^-1 and the temperature of drift tube for measurement was 89℃. Results: The linearity was in the range of 5 - 20μg for hosenkoside K and A, and the correlation coefficients ( n = 6) were 0. 9995 and 0. 9992 respectively; The average recoveries( n = 6) for hosenkoside K and A were 98.0% ( RSD = 1.3% ) and 98.0% ( RSD = 1.3% ) respectively. Conclusion:This method is accurate, reliable and reproducible for determination of hosenkoside K and A in the seeds of Impatiens balsamina.
