摘 要:目的:建立不同栽培品种玫瑰花药材的聚丙烯酰胺凝胶电泳(PAGE)鉴别方法。方法:蛋白质聚丙烯酰胺凝胶分析,电泳采用稳流控制,开始时电流15mA,当样品液面到达分离胶后,电流25mA。以泳道中各谱带所在位置Pos为横坐标,谱带透光率OD为纵坐标作图,绘制各谱带的透光率图并用EDAS软件分析谱带相似度。结果:采自山东平阴的丰花玫瑰与重瓣玫瑰含有的蛋白质种类相似,含量相似;采自北京妙峰山的金顶玫瑰与采自山东平阴的丰花玫瑰相似度较低;采自山东定陶的鸡心梅与丰花玫瑰相似度较低,蛋白质种类有一定差异;阴干品与烘干品蛋白质电泳谱带透光位置有较大差异。结论:玫瑰花蛋白质电泳的差异性表明了各品种间蛋白质种类的差异,可作为玫瑰花栽培品种鉴别的依据;加工温度对蛋白质影响很大。Objective: To develop a new method for identification of different cultivated species of Rosa rugosa Thunb. using polyacrylamide gel electrophoresis. Method:Analytical method has been established using polyaerylamide gel electrophoresis(PAGE). Electrophoresis, using steady flow control, starts with a beginning current of 15 mA, ends with a current of 25 mA when the sample liquid surface reaches separation plastic. Laning in the location of the various bands position for abscissa, transmittance bands OD for mapping ordinate, we draw the map of bands'light transmission rate and analyze banda similarity with EDAS software. Results:The results showed that the quantities and varieties of proteins in Fenghua Rosa which produced derived from Pingyin in Shandong province were similar to those of Chongban Rosa. Protein quantities and varieties derived from Beijing Miaofengshan Rosa rugosa Jinding and Shandong Pingyin Fenghua Rosa have a large difference. Protein varieties Shandong Dingtao Rosa chinertsis Jaca and Fenghua Rosa were also different greatly. Protein electrophoresis bands'transmittance location of drying samples and those dried in the shade differently greatly. Conclusion:The differences between of proteins indicated the species various breed of Rosa rugosa Thunb. can be used as a tool to identify different cultivated species of Rosa rugosa Thunb. In addition, the temperature in process and preparation also has significantly effects on the proteins in Rosa rugosa Thunb.
