猪巩膜胶原的纯化及胶原膜制备的研究

摘　　要：目的：研究猪巩膜胶原的提取纯化及眼用胶原膜的制备。方法：用盐析和ＤＥＡＥ纤维素３２柱层析纯化猪巩膜胶原，并用ＳＤＳＰＡＧＥ、凝胶扫描和氨基酸分析仪分析纯化的胶原成分及其氨基酸组成，经紫外照射使胶原交联，干燥成膜。结果：纯化胶原的电泳、凝胶扫描图和主要氨基酸组成与对照Ⅰ型胶原基本相同。结论：从猪巩膜提取纯化的胶原与胶原Ⅰ型类似。OBJECTIVE: To study extraction and purification of collagen from porcine sclera and preparation of collagen membrane. METHODS: Collagen of porcine sclera was extracted and purified by salting out and DEAE cellulose 32 colum chromatography. The protein components and amino acid composition were analysed with SDS PAGE. Gel scan and amino acid analyzer. The collagen was cross linked by ultraviolet irradiation and to form membrane by drying. RESULTS: Electrophoresis, gel scan figure and amino acid composition of the purified collagen of porcine sclera were mainly identical with reference collagen type Ⅰ. CONCLUSION: The collagen extracted and purified from porcine sclera is similar to collagen type Ⅰ. It is suitable for preparation of collagen membrane.
