摘 要:目的:观察通络救脑注射液对成年大鼠侧脑室室管膜下层Vimentin、胶质原纤维酸性蛋白(GFAP)阳性细胞的作用。方法:采用Wistar成年雄性大鼠,去除大鼠左侧大脑皮层血管,应用免疫组化方法观察去皮层血管对侧脑室室管膜下区(SVZ)神经干细胞增殖、分化和迁移的影响,以及通络救脑注射液的作用。结果:通过连续切片的免疫组化结果显示去皮层血管后侧脑室SVZ的增殖细胞核抗原(PCNA)、Vimentin、GFAP阳性细胞均明显增多,反应增强,表象改变显著。此种损伤刺激引起了相对静止的SVZ神经干细胞增殖、分化,给予通络救脑注射液后促进以上各种阳性细胞表达增多、增强。结论:提示通络救脑注射液可能通过上调内源性促神经生长因子的表达促进去皮层血管后成年大鼠SVZ神经干细胞增殖、分化。Objective: To study the effects of Tongluojiunao Injection on the Vimentin, GFAP positive cells located on the SVZ of adult rat. Methods: In this study, we use adult male Wistar rats, remove the left cortex vessels and adopt the immunohistochemistry (IHC) methods to explore the effects of devascularization of cortex on the proliferation, differentiation and migration of the SVZ NSCs, and in the meantime, the role of the Tongluojiunao Injection. Results: Through the IHC results of serial sections, we see that after the devascularization of cortex, the numbers of PCNA, Vimentin and GFAP positive cells increased greatly at lateral wall of SVZ. And their positive responses and morphologic changes are striking. Such trauma stimulates the relative static SVZ NSCs to proliferate and differentiate. After the administration of Tongluojiunao Injection, we can see the increasing of the expression of each positive cell. Conclusions: From this study we can make a speculation that Tongluojiunao Injection can prompt the SVZ NSCs of devascularization of cortex of rats to proliferate and differentiate through up-regulate the endogenous nerve growth factors.
