摘 要:目的:糖尿病是一种多系统代谢紊乱的疾病,其中以2型糖尿病为主要类型,目前对2型糖尿病的研究以提高胰岛素的增敏性,降低胰岛素抵抗为主要目的。方法:将SD大鼠分组喂以高脂饲料4周,造成2型糖尿病模型,给药组予人参皂苷Re、Rg1、Rb1分别给药4周,对照组予罗格列酮给药四周,取大鼠肝脏、肌肉、皮下脂肪组织,抽提蛋白,采用Western Blot混合上样,分别测肝脏组织中磷酯酰肌醇3激酶(PI3K),肌肉、脂肪组织中葡萄糖转运蛋白4(GLUT4)蛋白含量。结果:肝脏中PI3K的磷酸化表达Rg1组的蛋白表达要比Rb1组、Re组、高脂组、对照组和正常组有显著性差异,P<0.05;Rb1组、Re组与对照组的蛋白表达相近,但高于高脂组,接近正常组的蛋白表达。肌肉组织中Rb1组的GLUT4蛋白表达和Re组、Rg1组、高脂组、对照组和正常组有显著性差异,P<0.05;Re、Rg组、对照组高于高脂组的蛋白表达,接近于正常组的蛋白表达。脂肪组织中Rb1组GLUT4的蛋白表达和Re组、Rg1组、高脂组和对照组有显著性差异,P<0.05;接近于正常组的蛋白表达,Rg1组和Re组相近,对照组的蛋白和其他各组有显著性差异,P<0.05。结论:人参皂苷Rg1在肝脏组织中能促进PI3K的磷酸化,在肝脏组织中人参皂苷Rg1的抗胰岛素抵抗作用要优于Rb1组、Re组和对照组;而Rb1组、Re组和对照组在肝脏组织中仅有促进PI3K磷酸化的趋势,高于高脂组,说明人参皂苷在肝脏组织中有显著的促进PI3K磷酸化的作用。在肌肉组织中能促进GLUT4的表达,促进外周组织对葡萄糖的利用度,且Rb1、Rg1的这种作用明显优于西药对照组。人参皂苷可显著提高胰岛素受体(SRI)的信号通路,促进胰岛素受体的磷酸化,同时,可有效促进PI3K信号转导下游的葡萄糖摄取和转运,虽然在脂肪组织中表现出趋势,但在肌肉组织中表现出明显的促进作用,且这种作用较罗格列酮明显增加。Abstract: Objective:The diabetes is the disease that a kind of muti system supersession endangers the human health disorderlily and seriously. 2003 global diabetic's quantity is about 130 million people, among them more than 90% was Ⅱ type of Diabetes, and increase progressively at the speed of 1% every year, it is estimated that would increase sharply to 230 million until 2020 , has already become it following heart brain blood vessel disease and tumour, threaten the third major disease of the human health seriously. Study and compare ginsenoside Rb1 , Rg1 and Re function and mechanism that resist to the insulin. Methods: Divide the rats into 6 groups, feed through the high fat diet, cause the type Ⅱ diabetes model, after feeding for 4 weeks, observe the change of the weight of every group , fetch the muscle, adipose organize and liver organize, regard PI3K and GLUT4 as object albumen, observe expression of albumen to come with immune trace with western blot. Results: The weight of ginsenoside Re and Rgl increases and has significance differences with group Rbl by the look of weight, high with contrasting group's comparative weight closely ; In the liver organize taking PI3K as purpose albumen, the expression of the ginsenoside Rgl group with other groups have significance differences,P〈0.05. ginsenoside Re and normal group, ginsenoside Rgl have significance differences, ginsenoside Rb1 has the significance difference with ginsenoside Rg1. Taking GLUT4 as purpose albumen in the musculature, ginsenoside Rb1 and high fat group, contrast group, ginsenoside Re have significance differences, P〈0.05.In the adipose organize, ginsenoside Rb1, contrast group have significance differences,P〈0.05. Conclusion: Brief on the signal thorough fare where the ginsenoside can improve the insulin receptor (SRI) notably of ginseng, promote the phosphoric acid of the insulin receptor , meanwhile , can promote PI3K signal to transfer to the glucose which leads the low reaches absorbing and transporting effe
