摘 要:目的观察氟化物对体外培养乳鼠软骨细胞超微结构的影响。方法采用细胞培养的方法,原代培养昆明乳鼠软骨细胞,传第3代后按染氟剂量不同分为0(对照)、5、10、20、40mg/L组,染氟培养10d后,制备乳鼠软骨细胞超薄切片,透射电镜下观察软骨细胞的超微结构改变。结果电镜下5、10mg/L组软骨细胞增殖,双核细胞数量增多,细胞器发达,可见囊池扩张;20、40mg/L组细胞表面微绒毛减少,部分细胞染色质固缩,并凝结成块状,聚集在核膜周边,核基质密度下降,核膜迂回曲折,胞浆内线粒体空泡化,有些细胞胞浆内可见大量的囊泡,泡内含有电子致密物质;在40mg/L组中可见到凋亡细胞。结论高氟可以引起乳鼠软骨细胞超微结构的损伤。Objective To investigate the effects of fluoride of ultra - structure of chondrocyte of culture in vitro. Meth- ods The third generation of primary cultured chondrocytes were exposed to the concentration of 0 (control) ,5,10,20,40 mg/L fluoride for 10 days. Chondrocytes of newborn mouse were collected and ultra - thin sections were made and chondrocyte ultra - styucture was observed under eletronic microscope. Results After exposed to fluoride for 10 days, the proliferation was present in the chondrocyte of 5,10 mg/L groups and shrinked chromatine apoptosed chondrocytes were seen in 20,40 mg/L groups. Some mitochondria were seen swelling, the electronic density in this site increased, microvilli of chondroeyte decreased significantly. Conclusions Higher doses of fluoride can damage the ultra - structure of chondrocyte of newborn mouse.
