摘 要:目的探讨年龄对C57BL/6小鼠脑额叶(frontol,F)、背侧海马(dorsal hippocampus,Hd)、腹侧海马(ventral hippocampus,Hv)和嗅球(olfactory bulb,O)突触结合蛋白(Synaptotagmin I,SytI)量的影响。方法定量western blot测SytI及对照β-actin经软件处理用图像分析系统测印迹条带灰度值,SytI与β-actin灰度值比值SytI量用方差统计分析。结果正常脑衰老对额叶SytI量无显著影响(P>0.05)对背腹侧海马和嗅球SytI量均有显著影响(P<0.05)、老年SytI量高于成年。结论C57BL/6小鼠正常脑衰老背腹侧海马和嗅球SytI量随年龄增加示学习记忆相关背腹侧海马和嗅球在突触蛋白水平均有改变可能涉及年龄相关学习记忆能力减退发生;额叶SytI量无改变示额叶年龄相关改变可能后于海马和嗅球。Objective To explore the effects of age on the quanta of synaptotagmin I in frontol lobe,dorsal and ventral bippoeampus and olfactory bulb in C57BL/6 mice. Methods The quantitative Western Blot test was used to detect the expression of SytI and β - actin served as control in the different brain regions. The data were dealed with Photoshop CS software and Image - Pro Plus 5.0 system. All the variables were entered into SPSS 12.0 for Windows for statistical analyses. Results There was no age effect on Syt I quantum in the frontol lobe ( P 〉 0.05 ). Compared to adult mice, old mice had significant increase of the Syt I quanta in the dorsal and ventral hippocampus and olfactory bulb (P 〈0.05). Conclusion The Syt I quanta significantly increased with aging in the dorsal and ventral hippoeampus and olfactory bulb in C57BL/6 mice, but the Syt I quantum in the frontol lobe did not change with aging, suggesting the hippocampus and olfactory bulb more probably involed in the mechanism underlying the age - related decline of learning and memory ability.
