摘 要:对3个陆地棉材料子叶节进行离体培养,建立了高效的再生体系。其中,启动培养基为MSB+2.5 mg/L 6-BA+2.5 mg/LKT,芽分化培养基为MSB+0.3 mg/L 6-BA+0.3 mg/LKT,通过器官发生途径诱导形成丛生芽,芽伸长培养基MSB。以上培养基均含葡萄糖糖30 mg/L。此再生体系每个外植体可获得2.75个以上不定芽,嫁接移栽驯化后,成活率接近95%。In this paper, cotyledon nodes acted as the explants in vitro of three Gossypium hirsutum L. varieties were cultured, and multiple shoot were obtained by organogenesis method. An effective regenerative system was established in which cotyledon nodes were started in the start - up medium (MSB + 2.5 mg/L 6-BA + 2.5 mg/LKT), bud were differentiated in the differentiation medium ( MSB + 0. 3 mg/L 6-BA + 0.3 mg/LKT) and bud elongated in the bud elongation medium (MSB). Results showed that 30 mg/L glucose was served as carbon resource in all medium. Cotyledonary nodes produced maximum number of shoots (2.75 shoots/explant) and buds can be grafted. As the grafted seedlings were transplanted and domesticated, survival rate can reach nearly 95 %.
