清疣Ⅱ号方对SiHa细胞E7和PCNA蛋白表达的影响

摘　　要：目的探讨不同浓度清疣Ⅱ号方对体外培养SiHa细胞中E7和增殖细胞核抗原(PCNA)蛋白表达的影响。方法采用四甲基偶氮唑盐(MTT)还原法测定SiHa细胞的增殖抑制水平,采用间接免疫荧光技术观察各组细胞铺片中E7和PCNA蛋白的荧光表达情况。结果随药物浓度梯度的升高,各实验组细胞增殖活性逐渐降低,增殖抑制率逐渐增高。经中药清疣Ⅱ号方处理后,强表达E7和PCNA荧光的阳性细胞数减少,弱表达E7和PCNA荧光的阴性细胞数增加。E7和PCNA蛋白荧光表达的平均灰度随药物浓度梯度的升高逐渐增高,阳性单位随药物浓度梯度的升高逐渐减小。结论清疣Ⅱ号方能够有效下调E7和PCNA蛋白的表达,从而抑制体外培养的SiHa细胞增殖。这可能是其临床治疗尖锐湿疣,减少复发的药理作用机制之一。Objective To study the effects of Qingyou Ⅱdecoction on the E7 and proliferating cell nuclear antigen （PCNA） protein expression in SiHa cell. Methods M3T eolorimetrie assay was used to detect the proliferation inhabitation rate of SiHa cell. The E7 and PCNA protein expression in SiHa cell were measured by indrect immunofluorescene technology. Results A positive effect was shown along with the increasing of concentration, such as the proliferation inhabitation rate, the E7 and PCNA protein expression in SiHa cell. Conclution Qingyou Ⅱ decoction could obviously inhabited the proliferation level of SiHa cell,and down-regulated the protein expression of E7 and PCNA. It might be the effective therapy mechanism that Qingyou Ⅱ decoction could cure condyloma aeuminatum.
