摘 要:目的:建立亚临床肝性脑病大鼠模型,并探讨清开颗粒改善其学习记忆能力的机制。方法:采用四氯化碳(CC l4)高脂低蛋白复合因素造成的实验性肝硬化大鼠,对其进行动态的学习记忆测试及神经电生理测试直至出现亚临床肝性脑病。造模成功后分为中药治疗组、西药对照组及模型组,分别给予清开颗粒、乳果糖、生理盐水灌胃;2周后检测各组大鼠的Morris水迷津潜伏期成绩、肝功能、血清氨浓度和脑内氨浓度,及氨基酸神经递质的含量。结果:与正常组大鼠比较,至第10周时,造模后大鼠学习记忆的潜伏期明显延长;治疗组和对照组潜伏期均明显缩短(P<0.01)。治疗组和对照组血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、血氨、脑内氨浓度高于正常组(P<0.01),但明显低于模型组(P<0.01);治疗组的下降较对照组明显(P<0.01)。治疗组γ-氨基丁酸的含量低于模型组,谷氨酸与天冬氨酸的含量高于模型组,皆有统计学差异(P<0.01)。结论:清开颗粒可改善SHE大鼠的学习记忆能力,其作用机制可能与改善肝功能、降低氨浓度以及改善氨基酸神经递质有关。To establish the model of subclinical hepatic encephalopathy(SHE) in rats and investigate the mechanisms of "Qingkai Granule" in improving learning and memory. Methods: SHE rats were made by carbon tetrachloride (CC14 ) and high-fat and low-protein food. After successful modeling, the rats were randomized into treatment group, control group and model group, and respective- ly given "Qingkai Granule", lactulose and saline. Two weeks after treatments, the Morris water maze latency score, liver function, serum and brain anmmnia concentrations, and amino acid neurotransmitters were assayed. Results: Compared with normal rats, the latency of learning and memory in model group was significantly extended; the latency was shortened in treatment and control groups (P 〈 0.01 ). The serum ALT, AST, ammonia, ammonia concentration were higher in treatment and control groups, but lower than in model group( P 〈 0.01 ). γ-aminobutyric acid content was lower in treatment group than in model group ( P 〈 0.01 ), while glutamate and aspartyl acid contents were higher in treatment group than in control group (P 〈 0.01 ). Conclusion: "Qingkai Granule" can improve the learning and memory of SHE rats by improving liver function, lowering ammonia concentration and regulating amino acid neurotransmitters.
