摘 要:目的比较美国、荷兰、北京、上海4种血小板保存袋对手工制备浓缩血小板的保存效果。方法手工浓缩血小板无菌加入到4种保存袋于22℃保存,保存0、1、3、5、7d后分别取样检测血小板回收率、pH、体外聚集活性、血小板第3因子、第4因子有效性(PF4)及CD62P阳性表达率。结果随保存时间的延长4种保存袋血小板回收率均逐渐降低,但各保存袋之间无显著性差异(P>0.05);随保存时间延长pH均逐渐下降,血小板保存5d时各保存袋pH无显著性差异(P>0.05);保存7d时,北京保存袋pH(5.8±0.35)明显低于其他3种保存袋(6.68±0.13、6.88±0.13、6.96±0.11)(P<0.05)。随保存时间的延长各保存袋中的血小板聚集率均下降迅速,但4种保存袋间无显著性差异(P>0.05)。血小板保存5d时血小板第3因子(止血功能)无显著性差异(P>0.05);保存7d时美国(127.30±20.35)、北京(649.20±504.00)保存袋第3因子(纤维蛋白析出时间)比荷兰(93.38±7.51)、上海保存袋(71.10±4.52)明显延长(P<0.05)。保存7d血小板第4因子(释放功能)均无明显下降(P>0.05)。血小板保存5d时CD62P阳性表达率无显著性差异;保存7d时北京保存袋(56.63)高于其他3种保存袋(30.79、26.58、30.91)。结论北京、上海与美国、荷兰血小板保存袋相比,血小板保存5d时各检测指标接近,4种保存袋保存效果基本一致;但保存7d时北京保存袋其pH、血小板第3因子及CD62P阳性表达率指标比其他3种保存袋差。Objective To investigate the storage effect of four different storage bags (produced by America, Netherlands, Beijing, and Shanghai) for Platelet Concentrate. Methods Platelet Concentrate was divided into four different storage bags and stored in 22℃. The platelet count, pH, platelet aggregation activity, PF3AT, PF4AT and CD62P expression rate were measured on the dayO, dayl, day3, day5 and day7 during storage. Results There was no signifi- cant difference in platelet count among these four pletlet storage bags during storage of seven days( P 〈 0.05 ) ,although the platelet count all decreased gradually; There was no significant difference in pH among these four bags during storage of first five days ( P 〈 0.05 ) , however, on dayT, pH in Beijing storage bag (5.8 ± 0. 35 ) became significantly lower than in other bags(6.68 ± 0.13, 6.88 ± 0.13, 6.96 ± 0.11, respectively) ( P 〈 0.05 ) ; Platelet aggregation rate decreased rapidly in all four bags during storage, although there were no significant difference among four bags ( P 〈 0.05 ) ; There was no significant change of PF3 during first five days, while on dayT, American bag( 127.30 ±20.35 ) and Beijing bag (649.20 ±504.00) showed longer PF3 ( Fibrin precipitation time) than Netherlands bag ( 93.38± 7.51 ) and Shanghai Bag(71.10 ±4.52) ( P 〈0.05) ; There was little change of PF4 (release function) during seven days( P 〈0.05) ; There was no significant difference in the CD 6 2 P expression rate among the four bags during the first five days ( P 〈0.05) , but on day7, the CD62P expression rate of Beijing bag ( 56.63 ) was higher than other bags ( 30.79,26.58, 30.91). Conclusions During the first five days of storage, Beijing blood storage bag and Shanghai blood storage bag hove similar test indexes compared with American bag and Netherlands bag, which indicate that these four bags had similar storage effect for five days.
