摘  要:目的:研究CO2气腹对人宫颈癌CaSki细胞中survivin mRNA和蛋白表达的影响,探讨其促肿瘤生长的分子机制。方法:建立腹腔镜CO2气腹体外模型,以不同压力(0mmHg、7mmHg、14mmHg)的CO2气腹对人宫颈癌CaSki细胞处理不同时间(1h、2h、3h、4h),设未处理组为对照。采用RT-PCR法和免疫细胞化学法分别检测处理后各组细胞survivin mRNA和蛋白表达情况。结果:CO2气腹在一定的压力和作用时间范围上调CaSki细胞中survivin mRNA和蛋白的表达。结论:CO2气腹促肿瘤生长的效应可能是通过抑制肿瘤细胞的凋亡而实现的。Objective:To study the effect of CO2 pneumoperitoneum used in laparoscopy on the expressions of survivin mRNA and protein in human cervical carcinoma cell line (CaSki) . Methods:The CO2 pneumoperitoneum model was established in vitro. After CaSki cells were exposed to carbon dioxide pneumoperitoneum at 0mmHg, 7mmH scope or 14mmHg for 1h, 2h , 3h or 4h with the untreated group as control, the expressions of survivin mRNA and protein were examined by RT-PCR and immunocytochemistry. Results: Within certain pressure and exposure time, carbon dioxide pneumoperitoneums significantly upregulated the expressions of survivin mRNA and protein in CaSki cells. Conclusion:The promoting effect of carbon dioxide pneumoperitoneum on malignant cells growth is probably through inhibiting the apoptosis of tumor cells.
