MNP-NAG底物测定尿NAG方法的建立

摘　　要：应用自行合成的底物间甲氧基硝基乙烯基苯基 N-乙酰氨基β-D 葡萄糖苷(MNP-NAG测定尿 NAG 活性。以 NAG 酶作为标准,建立了底物冷冻干燥测定管试剂盒。色原 MNP 在碱性条件下呈红色,不受尿色干扰。用 MNP-NAG 的最适反应 pH4.6,批内变异系数 CV%<2%,批间 CV%<10%,与 PNP 终点法比较 r=0.98。测定了80例正常人尿 NAG 酶活性参考范围0.47～1.05U/mmolCr,并测定了部分肾脏病患者,具有重要的意义。This article introduces the measurement of urine NAG activity using substrate MNP-NAG.N-acetyl-β-D-glucosaminidase(MNP-NAG)was synthesized by our laboratory. The corresponding lyophilized substrate test tube kit was also set up with reference NAG in alkaline condition,the color-derived MNP appears red and is not interfered by the color of urine.The most optimal reaction pH of MNP-NAG is 4.6.The intraassay CV is less than 2 % and the inter-assay CV is less than 10%。The correlation coefficient is 0.98 comparing with PNP termination method.We measured the urine NAG activity of 80 normal persons and the reference value range form 0.47 to 1.05 U/mmol.Cr.
