摘 要:目的通过观察降脂酮对CCl4所致急性和慢性肝损伤大鼠肝组织、血清一氧化氮(NO)、一氧化氮合酶(NOS)水平的影响,探讨降脂酮对大鼠急、慢性化学性肝损伤保护作用的机制。方法大鼠分别腹腔注射50%CCl4花生油溶液1次(v/v)、皮下注射40%CCl4的花生油溶液5w,制作急、慢性化学性肝损伤模型;降脂酮灌胃后,测定大鼠肝组织、血清NO和NOS水平的变化。结果降脂酮可使CCl4所致急性肝损伤大鼠肝组织中NO和NOS水平明显降低,慢性肝损伤大鼠的血清和肝组织中NO和NOS水平无明显差异。结论降脂酮对急性肝损伤大鼠的保护作用可能与NO,NOS水平有关,对慢性肝损伤的保护作用可能与NO,NOS水平无关。Objective To evaluate the effect of Jiangzhi ketone on the expressions of NO, NOS of acute and chronical hepatic injury induced by CCl4 in rats. Methods The hepatic injury models were established by intraperitoneal injection 50% CCl4 for a single time or by subcutaneous injection 40% CCl4 in rats for 5 w. The rats were treated with Jiangzhi ketone at doses of 50, 100 and 200 mg/kg body weight. The content of NO, the activity of NOS in liver tissue were also observed. Results The levels of NO, NOS of acute hepatic injury rats treated with Jiangzhi ketone were significantly decreased, but there was no obvious change in NO and NOS levels in the rats with chronic hepatic injury. Conclusions Jiangzhi ketone plays protective role in acute hepatic injury rats by decreasing the expressions of NO and NOS but is not so in chronic hepatic injury rats.
