降脂复肝汤对高脂性脂肪肝模型大鼠肝脏保护作用及TNF-α表达的影响

摘　　要：目的观察降脂复肝汤对高脂饮食诱导的非酒精性脂肪肝大鼠肝组织病理学、肝功能、血脂及肿瘤坏死因子α(TNF-α)的影响。方法建立大鼠非酒精性脂肪肝模型。造模结束后,将造模组随机分为模型组、血脂康组和降脂复肝汤组,治疗12w后,通过HE染色、血清生化以及免疫组化检测,观察降脂复肝汤对肝组织病理学、血清酶学、血脂以及TNF-α的影响。结果血脂康组和降脂复肝汤组体重、肝重、肝指数、血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)及总胆固醇(TC)、甘油三酯(TG)低密度脂蛋白(LDL)均较模型组显著降低(P<0.01,P<0.05)。免疫组化法结果显示,降脂复肝汤组TNF-α的表达较模型组明显降低(P<0.01),而且,降脂复肝汤组比血脂康组作用明显(P<0.05)。结论降脂复肝汤能明显改善高脂饮食诱导的脂肪肝大鼠肝脏的病理变化、减轻肝组织脂肪变性,同时,也能降低脂肪肝变性肝组织TNF-α的表达,这可能是其治疗高脂饮食诱导的非酒精性脂肪肝的作用机制之一。Objective To observe the effects of Jiangzhi Fugan soup on the recovery of hepatic function and blood lipid and the expression of TNF-α of rats with non-alcohol fatty liver disease （NAFLD） and investigate its mechanism. Methods The SD rats were randomly divided into normal, model, Xuezhikang and Jiangzhi Fugan soup groups. Except the normal group,the other rats were fed with high fatty diet for 12 weeks to induce NAFLD. After the model were established, they were treated with Jiangzhi Fugan soup, Xuezhikang and NS respectively. 12 weeks later,the pathological alterations of liver tissues were examined by HE staining, the liver function was observed, the content of blood lipid was measured and the expression of TNF-α was detected by immunohistoehemistry. Results The serum levels of alanine transaminase （ALT）, aspartic transaminase （AST）, total cholesterol （TC）, triglyceride （TG）, low density lipid protein （LDL）, body weight, liver weight, hepatic index decreased in Jiangzhi Fugan soup and Xuezhikang groups （P 〈0. 01, P 〈0. 05） compared with model group. The expressions of TNF-α in in Jiangzhi Fugan soup and Xuezhikang groups were lower than that of model group（P〈0.05）, and the effect of Jiangzhi Fugan soup was more obvious （P〈0.05）. Conclusions Jiangzhi Fugan soup can improve the change of hepatic pathology and fatty degeneration and decreas the expression of TNF-α, which may be one of mechanisms of anti-NAFLD induced by high fatty diet.
