摘  要:目的目的观察红花对结扎单侧输尿管诱导的肾间质纤维化实验大鼠肾小管上皮细胞表型转化的抑制作用。方法结扎单侧输尿管方式复制肾间质纤维化实验动物模型,分别给以红花4.0g·kg-1·d-1、缬沙坦10mg·kg-1·d-1经口灌服,采用免疫组化、原位杂交方法检测大鼠肾脏肾小管上皮细胞表型转化标志物α-平滑肌肌动蛋白(α-SMA)及Ⅲ型胶原的表达。结果单侧输尿管结扎大鼠肾脏α-SMA动蛋白和Ⅲ型胶原的阳性面积及积分光密度较假手术组明显增强,红花及缬沙坦可显著性抑制其表达。结论红花可以通过抑制肾小管上皮细胞的表型转化拮抗肾间质纤维化。Objective To observe the effect of Safflower on tubular epithelial-myofibroblast transdifferentiation in renal interstitial fi- brosis rats induced with unilateral ureteral obstruction (UUO). Methods Renal interstitial fibrosis rats induced with UUO were given orally Safflower(4.0g·kg^-1·d^-1) and Valsartan (10g·kg^-1·d^-1) respectively. The marker of epithelial-myofibroblast transdifferentiation α-smooth muscular aetin(α-SMA) and collagen Ⅲ were examined by immunohistochemical method and in situ hybridization. Results The positive area and density of α-SMA and collagen Ⅲ in UUO rats increased markedly than those in sham operation group and could be inhibited by Safflower and Valsartan. Conclusions Safflower can antagonize renal interstitial fibrosis by inhibition of epithelial-myofibroblast transdif- ferentiation.
