摘  要:目的探讨辛伐他汀对糖尿病大鼠肾脏结缔组织生长因子(CTGF)表达和排泄的影响。方法建立糖尿病大鼠模型,分为对照组、模型组、辛伐他汀(20mg/kg·d)组,检测各组第1、4、6、8周血糖、尿CTGF排泄率(ELISA法),进行组间差异比较;第8周应用免疫组化检测肾皮质CTGF蛋白表达。结果第4、6、8周糖尿病大鼠治疗组及糖尿病模型组尿CTGF排泄率及第8周肾皮质CTGF蛋白表达水平较正常组明显升高,治疗组上述指标均较模型组低(P<0.01)。结论辛伐他汀可抑制肾脏CTGF表达和排泄,这可能与其保护肾脏有关。Objective To study the effect of Simvastatin on expression and excretion of connective tissue growth factor (CTGF) in renal tissue and urinary of diabetic nephropathy rats. Method Three groups of rats were studied: normal group ( n = 8 ), STZ-induced diabetic model group ( n = 8 ), diabetes Simvastatin-treatment group ( n = 8 ). Blood glucose and urinary CTGF were measured at the 1 st,4th,6th and 8th week,as well as the expressions of CT-GF protein ( by histochemical staining) at the 8th week by ELISA. Results The urinary excretion rates of CTGF at 4th,6th,8th weeks and the expressions of CTGF protein in renal cortex at 8th weeks significantly increased in STZ-induced model group compared with those in normal group (P 〈 0. 01 ) ; Simvastatin-treatment decreased the urinary excretion rates of CTGF and the expressions of CTGF protein in renal cortex compared with those in model group significantly (P 〈 0.01 ). Conclusion Simvastatin can suppress the expressions of CTGF protein in renal cortex, as well as the urinary excretion rates of CTGF in diabetic rats, which suggest that the renal protective effects of Simvastatin maybe partly due to their effects in down-regulating the CTGF production of renal tissue.
