大鼠坐骨神经-20℃川芎嗪玻璃化液保存对异体移植后神经再生的影响

摘　　要：目的:探讨不同时间段-20℃川芎嗪玻璃化液保存大鼠异体坐骨神经对异体移植后神经再生的影响.方法:-20℃川芎嗪玻璃化液(200mg/L川芎嗪)(A组)保存成年SD雄性大鼠坐骨神经,时间分别为1,2wk,然后选择成年Wistar雄性大鼠坐骨神经缺损模型作为受体,神经段复温后移植给受体,术后进行大体观察以及移植神经的大体、电生理、荧光逆行示踪、运动终板等检查;并分别与-20℃玻璃化液(不含川芎嗪)(B组)和-196℃川芎嗪玻璃化液(200mg/L川芎嗪)(C组)保存的坐骨神经进行比较.结果:保存1wk时-20℃川芎嗪玻璃化液组与-196℃川芎嗪玻璃化液组再生效果无差异,与-20℃玻璃化液组的差异有显著性(P<0.01);保存2wk以后3组中以-196℃川芎嗪玻璃化液组再生效果最好,-20℃川芎嗪玻璃化液组其次,-20℃玻璃化液组最差.结论:玻璃化保存液中加入川芎嗪可提高大鼠同种异体神经的保存温度值,-20℃时可保存大鼠周围神经1wk.AIM： To study the regeneration of sciatic nerves allografts preserved in the vitrification solution containing tetramethylpyrazine at -20℃ within different time period. METHODS： Sciatic nerves taken from male SD rats were preserved in vitrification solution containing 200 mg/L tetramethylpyrazine at -20℃ for 1-2 weeks respectively （Group A ）. The segments of these nerves were then transplanted to the corresponding Wistar rats with part of sciatic nerve defects. The allografts were examined with the gross observation, electrophysiology, immunofluorescence and counts of motor end-plate postoperatively. The results were compared respectively between Group A, Group B （vitrification solution at - 20℃ ） and Group C （ vitrification solution containing tetramethylpyrazine at - 196℃ ）. RESULTS ： No statistical difference was found between Group A and Group C, but significantly different was seen between Group A and Group B （ P 〈 0.01 ） for the regeneration of allografts preserved for 1 week. After the 2-week preservation, the regeneration was the best in Group C and the worst in Group B. CONCLUSION： The preserved temperature of peripheral nerve allografts can be raised by adding tetramethylpyrazine to the vitrification solution, in which peripheral nerve allografts can be preserved at -20℃ for 1 week.
