摘 要:目的:探讨雌二醇(E2)对去势大鼠局灶性脑缺血再灌注后的保护作用及其对凋亡抑制基因bcl-2的影响.方法:建立雌性Wistar大鼠脑缺血再灌注模型(MACO),分为4组,WT去势大鼠安慰剂组(A组)、苯甲酸雌二醇组(B组)、ERKO去势大鼠安慰剂组(C组)、苯甲酸雌二醇组(D组).采用放射免疫分析、激光多普勒血流(LDF)仪、免疫组化和RT-PCRC等检测血浆E2水平、脑血流量的变化、促血管生成素-1(Ang-1)mRNA的表达、调亡指数以及bcl-2表达.结果:经E2处理的雌性野生(WT)和雌激素α受体基因敲除(ERKO)去势大鼠,其脑缺血再灌注后脑血流的变化幅度、脑组织中Ang-1mRNA表达和bcl-2的表达均高于安慰剂对照组(P<0.01).结论:E2介导的Ang-1mRNA和bcl-2表达的增加及其对微血管结构的稳定作用是脑缺性再灌注损伤修复的重要机制.AIM : To investigate the protective effect of estradiol on angiopoietin-1 mRNA ( Ang-1 mRNA ) and bcl-2 gene expression in castrated rats with focal cerebral ischemia reperfusion. METHODS: Cerebral ischemia reperfusion models were established in female Wistar rats and the rats were divided into WT castrated rat placebo group ( A group ) , estradiol benzoate group ( B group) , ERKO castrated rat placebo group( C group) and estradiol benzoate group( D group). The plasma estradiol levels, changes in cerebral blood flow, Ang-1 mRNA, apoptosis and bcl-2 gene expressions were determined respectively by radio-immunity analysis, laser Doppler flowmetry (LDF), immunohistochemical stain and reverse transcription polymerase chain reaction (RT-PCR). RESULTS: The plasma estradiol levels in WT and ERKO castrated rats treated with estradiol were higher than those in respective control groups( P 〈 0.01 ). In group of WT MCAO castrated rats treated with estradiol, the change in cerebral blood flow after cerebral ischemia repeffusion, the Ang-1 mRNA expression and bcl-2 gene expression in brain tissues were higher than those for their respective control (P 〈 0.01 ). CONCLUSION: The increased expression of Ang-1 mRNA and bcl-2 gene expression mediated by estradiol and their stabilizing effect on capillary vessels are the important repair mechanism of cerebral ischemia reperfusion injury.
