摘 要:研究两级泡沫分离提取多粘菌素E的工艺。考察溶液初始装液量、分离时间、溶液pH值、氯化钠浓度等操作参数对第一级分离过程的影响。结果表明:第一级分离在最佳工艺为装液量1000mL,气速0.10m3/h,pH为5.0,NaCl浓度5g/L,分离时间2.5h时,多粘菌素E收率为90.00%,富集比为4.00。通过正交试验对第二级分离参数优化,其最佳工艺为气速0.05m3/h,装液量600mL,pH为5.0,分离时间2h时,第二级分离回收率为75.62%,富集比4.57。A technology of two-stage foam fractionation for recovery of Polymyxin E was studied. The effects of operation variables of volume of initial solution, separation time, pH, NaCl concentration on the first stage separation process were investigated. The results showed that when the optimum conditions of the first stage separation were liquid volumn 1 000 mL, gas velocity 0.10 m^3/h,pH 5.0,concentration of NaCl 5 g/L and separation time 2.5 h, the recovery and enrichment ratio of Polymyxin E were 90 % and 4.0, respectively. The optimum conditions of the secondary stage separation were gas velocity 0.04 m^3/h, volume of liquid 600 mL, pH 5.0, NaCl 5 g/L and separation time 2 h. By orthogonal experiments design, the recovery and enrichment ratio of Polymyxin E of the secondary stage separation were 75.62 % and 4.57 at the optimum conditions.
